I really like the idea of Iago as a puppeteer- he knows what
strings to pull on all the other characters to set them off
against each other. One of Othello's strings is that he is
an outsider- if he was led to believe young venetian women
typically fell for young venetian men, and liked to spite their
fathers, well that's something.
There does seem to be something else though- the whole hankie
thing bears a lot of significance in the idea of time. Iago
claims he saw Cassio wipe his beard earlier in the day with the
cloth Othello gave to Desdemona. Othello decides this is proof
enough- when only minutes ago (maybe about 100 text lines up?),
his wife used the handkerchief to bind his head! So I'd be tempted
to say there is a jealous nature in Othello for him to jump to
such conclusions so quickly.
